The AAV-shRNA Selection Kit is designed for efficient selection of shRNAs. Each AAV-shRNA Selection kit includes 4 pre-designed shRNA constructs targeting your gene of interest (GOI), 1 scramble shRNA as control, and a GOI-mCherry fusion reporter plasmid. All plasmids are carried two AAV ITRs, after selection,  the constructs can directly used to package AAV vectors.

Kit Components

  1. Pre-designed shRNA Plasmids
    • 4 target-specific shRNA constructs  (eg. pAAVtri-U6-shRNA-CMV-EGFP)
    • 1 scrambled control shRNA
  2. Reporter Plasmid (pAAVtri-CMV-GOI-mCherry)
    • GOI-mCherry fusion for real-time knockdown assessment
  3. Backbone Flexibility
    • Reporter expression: CMV or EF1α promoters
    • shRNA expression: U6 or H1 promoters
  4. Dual-Reporter System
    • EGFP: Visualize transfection efficiency
    • mCherry: Quantify gene silencing efficacy

Workflow:

  1. Co-transfection: Deliver shRNA plasmids + GOI-mCherry plasmid into cells (e.g., HEK293T).
  2. Screening: Analyze mCherry/EGFP ratios (flow cytometry or qPCR) to identify optimal shRNAs.
  3. AAV Packaging: Selected shRNAs can be packaged into AAV vectors (individual or pooled purification).
