AAV Plasmids for AAV Production

To address AAV production challenges, AAVnerGene has developed innovative technologies aimed at improving traditional triple-plasmid AAV production systems packaging efficiency:

  • AAVtri (Triple-Plasmid System)
    • mini-pHelper-1.0 – A compact adenoviral helper plasmid (8.4 kb) delivering E2A, E4orf6, and VA RNA for improved packaging efficiency over traditional Ad helper plasmids.
    • pRCap-AAV – An optimized AAV helper plasmid featuring a modified Rep expression cassette, enhancing packaging efficiency compared to standard AAV2 Rep.
    • pAAVtri – Regular AAV cis plasmid carrying two ITRs and GOI expression cassette.

Building on the mini-pHelper-1.0 and pRcap, AAVnerGene has introduced two advanced systems:

  • AAVone® (Single-Plasmid System)
    • AAVone1.0: The first generation of AAVone [Yang et al., 2025].
      • Combines all essential components—Ad helper genes (E2A, E4orf6, VA RNA), AAV helper genes (Rep/Cap), and the AAV vector genome—into a single plasmid (pAAVone
      • Achieves 2-4 fold yield vs AAVtri.
    • AAVone2.1: The second generation of AAVone system
      • Achieves full-particle ratio at harvest up to 80%.
      • Achieves high-yield AAV production (AAV9 >5 × 10¹⁵ vg/L) in HEK293 cells
  • AAVdual® (Two-Plasmid System)
    • Integrates the AAV genome and transgene into the optimized mini-pHelper-1.0 backbone (pAAVdual) [Yang et al., 2025].
    • Balances high AAV productivity with AAV capsid flexibility.

These systems significantly streamline AAV production, reducing complexity and improving scalability for both research and clinical applications.
