mini-pHelper-AAV-DJ is a compact, optimized plasmid designed to produce AAV-DJ empty particles or AAV-DJ vectors. It integrates AAV-DJ Rep and AAV-DJ Cap genes into our mini-pHelper-1.0 backbone. Thus, it not only provides adenoviral helper functions (E2A, E4orf6, VA RNA), but also AAV helper functions (Rep and AAV-DJ). When transfected into host cells alone, it facilitates the generation of truly-empty AAV-DJ particles without any genetic materials inside of capsids. Co-transfection mini-pHelper-AAV-DJ with pAAVtri-GOI enables high-efficiency production of recombinant AAV-DJ vectors carrying your gene of interest (GOI).

- Version V1: Combining Ad helper with GOI: V1, exemplified by AAVnerGene’s AAVdual system, stands out for its ability to maintain both packaging efficiency and flexibility. This version allows researchers to work with various AAV serotypes while ensuring efficient AAV vector packaging. This balance is particularly beneficial for projects involving diverse gene therapy or vector biology applications.
- Version V2: Combining Ad helper with AAV helper: V2, such as the pDG and pQT systems, offers flexibility in producing different AAV serotypes, making it useful for a wide range of research applications. AAVnerGene’s mini-hepler-AAVs are pDG like plasmids, but more compact in size.
- Version V3: Combining GOI with AAV helper: V3, such as the pOXB system, may provide increased AAV packaging efficiency, which is critical for maximizing the yield of properly packaged AAV vectors. However, it sacrifices flexibility by linking the GOI with the AAV helper plasmid, making it more challenging to switch and customize the GOI.
