The EFS promoter, a truncated version of the human EF1α regulatory sequence, is engineered to deliver dependable, continuous gene expression in a wide range of cell types while occupying minimal vector space. By condensing the EF1α promoter into a compact format, it preserves strong transcriptional output yet remains far smaller than the full‑length version, making it well suited for applications with limited cargo capacity.

The Ado1.0 is a GPCR‑based EGFP fluorescent sensor designed to visualize extracellular adenosine dynamics with high sensitivity and cellular specificity. It is engineered by integrating a circularly permuted fluorescent protein into the human A2A adenosine receptor so that ligand‑induced structural shifts produce measurable fluorescence changes.

DIO‑Ado1.0 is a Cre‑dependent version of the medium‑affinity GRAB adenosine sensor, engineered so that expression occurs only in cells expressing Cre recombinase. It uses the EFS promoter to drive reliable, constitutive expression once activated, enabling targeted measurement of extracellular adenosine dynamics in defined neuronal populations.

To ensure high-quality and pure products, AAVnerGene uses various assays to assess the quality and purity of their AAV vectors:

  • Gene specific qPCR Titer: Target the specific transgenes or common used elements, such as WPRE, CMV, PolyA, but not ITR.
  • Full/Empty Particle Ratio: Ensure all the premade AAVs has >85% full particles by Mass Photometry.
  • Endotoxin Testing: Ensure safety with endotoxin level <1U/1e13VG.
  • SDS-PAGE Gel: Assess the purity of the AAV viruses is great than 95%.
  • Infectivity Assay: Evaluate the efficiency of AAV viruses in transducing target cells if they have reporters.
