AAV Capsid Kits

AAV vectors are powerful tools for gene delivery and gene therapy. With hundreds of identified natural AAV serotypes and an increasing number of engineered variants, researchers can select optimal capsids for specific cell types and tissues—an essential factor for effective gene therapies. However, achieving this selection has proven challenging. The common approach involves testing AAV variants side by side individually, which complicates the generation, control, and handling of hundreds of AAV vectors in every lab or company. This limitation reduces the likelihood of identifying superior AAV variants.

To address this challenge, AAVnerGene offers two types of AAV Capsid Kits designed to help customers efficiently evaluate and select various AAV serotypes and variants tailored to their specific applications.

  • AAV Capsid Barcode Kits are advanced AAV capsid selection tools built on our ATHENA-I These kits systematically evaluate a group of AAV serotypes or variants using Barcode-Seq technology and next-generation sequencing (NGS). In the AAV Capsid Barcode Kits, each capsid variant is assigned 3 distinct DNA barcodes (BCs). By comparing the DNA or RNA levels of these barcodes through NGS, researchers can identify which AAV capsid variant is the most efficient for their specific applications.
  • AAV Serotype Testing Kits are designed to quickly and efficiently identify a select group of commonly used AAV serotypes by measuring the expression levels of fluorescent proteins (EGFP, mCherry, TdTomato) or luciferase (Gluc, Rluc, Fluc, Cluc). This method is particularly useful when researchers have a clear understanding of the cell lines or tissues they intend to target and seek a straightforward way to narrow down their choices to a smaller subset of AAV serotypes.

AAV Capsid Barcode Kits

AAVnerGene offers both common and tissue-specific AAV Capsid Barcode Kits, as well as custom development services.  Customers can create kits with fewer than 25 capsids or develop AAV Capsid BC-Libraries with more than 25 capsids. Each kit can be tailored to include any AAV serotypes and variants, with the added flexibility to customize reporters and promoters to meet specific needs. The AAV Capsid Barcode Kits – Common include 15 frequently used AAV serotypes, specifically: AAV1, AAV2, AAV3B, AAV5, AAV6, AAV7, AAV8, AAV9, AAVrh.10, AAV11, AAV12, AAV13, AAVrh.74, AAV-DJ, and AAV2-Retro. Researchers can choose from various reporter systems, such as AAV-CAG-EGFP, AAV-CMV-mCherry, and scAAV-CMV-mCherry.

AAV Barcode plasmids are used to generate AAV Capsid Barcode Kits and AAV Capsid BC-Libraries. Each plasmid contains a unique DNA barcode, allowing for the identification and tracking of individual AAV capsid in the kits or libraries.

AAV Serotype Testing Kits

In this category, 15 most used AAV serotypes (AAV1, AAV2, AAV3B, AAV5, AAV6, AAV7, AAV8, AAV9, AAV11, AAV12, AAV13, AAV-DJ, AAVrh.10, AAVrh.74, and AAV2-Retro) are included in each kit. The vectors carry fluorescent protein (EGFP, mCherry, TdTomato) or reporter luciferase (Fluc, Rluc, Cluc, Gluc), which is driven by a promoter (CAG, CMV, EF1a or hSyn). The kits are pretended to provide useful tools for customers, with minimum cost, in testing AAV transduction efficiency, testing tissue specific tropism related to serotypes, and using as negative control to examine whether the observed biological effects come from specific transgenes.
