AAVone system

AAVone System

The AAVone System integrates all necessary components for AAV vector production into a single compact plasmid (pAAVone) with a 13 kb oversized backbone. This plasmid includes all adenoviral helper genes (E2A, E4orf6, and VA RNA), AAV helper genes (Rep and Cap), and the AAV vector genome. AAV vectors can be generated simply, efficiently, and consistently by transfecting this single plasmid into host cells. The AAVone System has demonstrated impressive results, achieving unpurified yields exceeding 1×10^15 viral genomes (VGs) per liter in suspension-cultured HEK-293T cells for most AAV serotypes. This yield is 2 to 4 fold higher than that achieved with the original triple-plasmid transfection system. For AAV9, AAVone can reach 5e12 VG/ml in suspension cultured HEK 293 cells. In addition to increasing yield, the AAVone System reduces the number of plasmids required from 3 to1, as well as decreasing the total plasmid amounts needed for each production run. This streamlined approach not only enhances efficiency but also simplifies the manufacturing process for AAV vectors.

AAVdual System

AAVdual System

In the AAVdual System, the AAV genome with transgene is assembled into our novel mini-pHelper to form the pAAVdual plasmid. AAV vectors can be flexibly and efficiently generated by co-transfection of pAAVdual plasmids with the regular AAV helper plasmids (pRCap) into host cells.
The AAVdual System:
           ---- Allows for modular design and flexibility in constructing AAV vectors. 
           ---- Exhibits high efficiency in generating AAV vectors.
           ---- Streamlines the transfection process by reducing the number of plasmids. 

AAVtri System

AAVtri System

In AAVnerGene's AAVtri System, the original pHelper plasmid has been replaced with a smaller version known as mini-pHelper (8.4 kb). This modification retains the traditional triple-plasmid approach, where AAV vectors are produced by co-transfecting the mini-pHelper with the pRCap and pAAVtri plasmids into host cells. The reduced size of the mini-pHelper enhances the efficiency of transfection, increase AAV productivity, resulting in lower overall production costs. All the AAVdual and AAVone systems are developed based on this mini-pHelper.
