AAVdual System
The AAVdual® System is one of dual-plasmid AAV production systems, which was developed based on our novel mini-pHelper-1.0 plasmid. In this system, the AAV genome carrying the transgene is cloned into the mini-pHelper-1.0, resulting in pAAVdual plasmids. AAV vectors are then generated by co-transfecting pAAVdual plasmids with a standard AAV helper (pRCap) plasmid. This system provides high flexibility and enhances AAV packaging efficiency in vector production.
The AAVdual system streamlines the AAV production process, offering several key advantages:
- Streamline the AAV production process. Compared with traditional triple plasmid system, it reduced plasmid number from 3 to 2, by combining the mini-pHelper with pAAVtri-GOI:
• pAAVdual Plasmid: Contains the gene of interest (GOI) flanked by inverted terminal repeats (ITRs) for AAV replication and packaging as well as Ad helper genes (E2A, E4orf6 and VA RNA).
• pRCap: AAV Rep-Cap Plasmid encodes the necessary proteins for AAV genome replication and capsid assembly. - High Packaging Efficiency: Demonstrating a significant improvement of 100% to 250% across multiple serotypes when compared to the AAVtri system.
- Streamlined Workflow: With fewer plasmids to manage, the AAVdual system simplifies the workflow, saving time and resources during production.
- Flexibility in Serotype Production: Easily generate AAV vectors with different serotype specificities by selecting appropriate Cap genes, all within the same in AAVdual system framework.
- Compact Plasmid Size: The pAAVdual and pAAVdual-sc plasmids measure only 11±2 kb, making them easier to handle and produce without significantly affecting plasmid yield. This compact size also facilitates cloning procedures.

- All the plasmids are kanamycin resistance (KanR).
- A Material Transfer Agreement (MTA) is required for academic use of all mini-pHelper–based plasmids, including pAAVdual plasmids, and a license agreement is required for industrial use.
