AAV vectors are powerful tools for gene delivery and gene therapy. Hundreds of identified natural AAV serotypes and increasing number of engineered variants allow us to find better capsids for each cell type and tissue, which is vital for effective gene therapies. However, there has been no good method to achieve that. Currently, the common way to compare AAV variants is to test them side-by-side individually, whereas it is difficult to generate, control and handle hundreds of AAV vectors in every lab or company while limited candidates decrease the chance to find better AAV variants.
AAVnerGene offers two solutions to help customers efficiently compare and select different AAV serotypes and variants for specific applications.
- AAV Serotype Testing Kits: These kits are designed to quickly and efficiently identify a limited set of used AAV serotypes that are suitable for specific cell lines or tissues. This method is likely useful when you have a clear understanding of the cell lines or tissues you intend to target and want a simple way to narrow down your choices to a smaller subset of AAV serotypes. The testing process probably involves exposing the cells or tissues to various AAV serotypes and observing how effectively they transduce the target cells. This can provide you with a preliminary indication of which AAV serotypes are most effective for your specific experimental setup.
- AAV Capsid Library(I): The AAV capsid library(I) is a known AAV capsid library designed to systematically evaluate a larger number of AAV serotypes or variants using next-generation sequencing (NGS) methods and DNA barcode technologies. This method is likely more comprehensive and is suited for cases where you want to explore a broader range of AAV serotypes and variants. NGS allows for high-throughput sequencing of genetic material, enabling you to analyze the transduction efficiency, tropism, and other characteristics of multiple AAV serotypes simultaneously. This approach provides a more detailed understanding of how different serotypes interact with your target cells or tissues and can help you make more informed decisions about which AAV serotypes to use.
AAV Serotype Testing Kits
In this category, 16 most used AAV serotypes (AAV1, AAV2, AAV3B, AAV4-Y729F, AAV5, AAV6, AAV7, AAV8, AAV9, AAV11, AAV12, AAV13, AAV-DJ, AAVrh.10, AAVrh74, AAV2-retro) are included in each kit. The vectors carry fluorescent protein (EGFP, mCherry, TdTomato) or reporter luciferase, which is driven by a promoter(CAG, CMV, EF1a or hSyn). The kits are pretended to provide useful tools for customers, with minimum cost, in testing AAV transduction efficiency, testing tissue specific tropism related to serotypes, and using as negative control to examine whether the observed biological effects come from specific transgenes.
AAV serotype testing kits can be purchased directly as they are, or they can be customized based on the specific needs of the customer. We also provide custom AAV serotype testing kits. Customers can add any AAV serotypes and variants into the kits. Both reporter and promotor also can be customized basing on the customer needs
